Canadian researchers at the Montreal Clinical Research Institute have discovered that two protein complexes, known as Polycomb complexes (PRC1 and PRC2), act as "genetic brakes" by switching off genes at critical moments during limb development in embryos. When these molecular systems function properly, they allow initial developmental genes to be silenced so that subsequent genetic programs can take over; however, disrupting both systems simultaneously causes early genes to remain active and severely compromises normal limb formation. The findings, published in PNAS, reveal a fundamental principle of embryonic development: cells must not only activate the correct genes but also maintain precise control over which genes remain switched off to prevent congenital abnormalities.
